Puzzles like this have been around for a long time. The trick to solving them is to remember that one square can be made up of smaller squares. Even though they're different sizes, you need to count them all.
You'll have an easier time getting the right answer if you start with the smallest ones and count all of them, then move onto the next size and count those, until you get to the largest size.
Remember, all squares are rectangles, but not all rectangles are squares. It's only a real square if the sides are the same length. If someone tells you to count rectangles, they're forgetting basic geometry.
People who say 42 are making a reference to The Hitchiker's Guide to the Galaxy, wherein "the answer to life, the universe, and everything" is the number 42 (the question to life, the universe, and everything remains ambiguous).
Cudos to the clever rat who first said the solution was 102,400 which is the number of pixels on the default image. Technically, pixels are squares, though that's probably not what the puzzle-maker intended.
